A rabbi was asked this question and said that sometimes you can know by your last name, but most of the time you don’t know. His last name is from the tribe of Levi, and he has been fitted as a priest for the Temple. Sometimes you know by tradition. To say that you matched the DNA of a tribe, you would have to have the original DNA to determine if yours matches the Levitical tribe.
Regarding the Millennial Temple priest, Jesus Christ will set that up, and King David will rule and reign (Ezekiel 42-48) and reinstitute the Temple practices. Jesus Christ will know who will be qualified to do the priestly duties in the Millennial temple.
